我國研發流感新藥「零流感」（Tamiphosphor）有重大進展！中央研究院基因體中心運用「溴苯」（bromobenzene）作為起始物，將製程從四十五天縮短至十四天、總產率提高兩倍，目前已進行動物實驗。研發團隊預計五到十年內，市面上就會有真正「made in Taiwan」的抗流感藥物。


此研究論文，也獲得國際知名期刊《Angewandte Chemie》以「熱門論文」方式刊登。


中研院研發 效果比克流感好


「克流感」是目前治療Ｈ５Ｎ１、Ｈ１Ｎ１禽流感的唯一口服藥，但隱憂不小。挪威統計，流感病毒對「克流感」有二○％的機率產生抗藥性，且「克流感」原料來自八角萃取出的苯草酸（shikimic acid），收集需投入大量人力，製造過程中還需使用「疊氮鈉」（sodium azide），容易引起爆炸反應，羅氏公司將工廠設置於北韓，而且，最具危險性的步驟必須在地底深處進行。


中研院去年研發出新藥「零流感」，與「克流感」同屬流感病毒的神經醯胺抑制劑，但在酵素活性、保護細胞等結果上，都比「克流感」效果更好。且實驗小鼠服用「零流感」後的存活率較服用「克流感」高，體重恢復情形較明顯。


製程從四十五天縮短為十四天


然而，先前使用木醣（xylitol）作原料，製程需四十五天，效率較低。中研院改以溴苯作為起始物，一步一步加入其他官能基，發展出一條截然不同的合成方法，較舊方法減少八個步驟，製程從四十五天縮短為十四天，同時，可直接利用大腸桿菌工廠大量發酵的產品，許多中間產物均可結晶，減少層析的純化步驟，製造方式更為經濟。而「溴苯」為石化原料，一般大學二年級學過有機實驗的學生都能合成，比八角、木醣更容易取得。


該重大突破使台灣抗流感藥物更具商業價值，獲得國際知名期刊《Angewandte Chemie》以「熱門論文」方式刊登，研發團隊包括中研院長翁啟惠、研究員方俊民及博士後研究員謝俊結。


方俊民表示，目前已有藥廠有意願投入資金，保守估計最快五年內就能完成人體實驗，進入量產上市階段，為我國防範禽流感、人類流感病毒，提供本土開發的藥品，治療更多的病患。
